Unlike classical uncouplers such as FCCP, BAM15 demonstrates selectivity for mitochondrial membranes without depolarizing plasma membranes. Mechanism of Action: BAM15 functions as a lipophilic weak acid protonophore that transports protons into the mitochondrial matrix via a pathway independent of ATP synthase, thereby uncoupling nutrient oxidation from ATP production. EC₅₀ = 270 nM in L6 myoblast mitochondria. Critically, BAM15 stimulates higher maximum mitochondrial respiration rates than FCCP while preserving plasma membrane potential and demonstrating lower cytotoxicity. Does not require adenine nucleotide translocase (ANT) for activity.
